Abstract

Soil salinity is the main and pervasive problem in many countries like Iran. Complexity of salinity tolerance and its multi-gene heredity are important factors, which caused problems in providing salt-tolerance cultivars. Tritipyrum as one of amphiploid cultivar, was created from crosses between wheat and wild wheat species, genus thinopyrum. Tritipayrum obtained salt-tolerance genes from thinopyrum as paternal plant with more than 350 mM salt tolerance capacity. In this study application of 80 primers to identify Eb genome in Tritipyrum lines showed no specific fragment corresponded to the primers. In addition identification of Eb genome chromosomes using markers was completely random and non-repeatable. Therefore, OPF03 primer as a specific marker of Eb genome was identified based on the experiments performed by other researchers. Two primers called F1R1 and F2R2 were designed based on the deposited sequence in the gene bank. The primers could amplify two fragments of 500bp and 1272bp in lines Tritipyrum and Thinopyrum bessarabicum, however the bands were not found in Chinese spring wheat as a salt-sensitive varietie. The amplified fragments were sequenced and result showed 94%-97% homology with Eb genome sequence. As a conclusion in Tritipyrum lines the amplified fragments could be inherited from paternal plant (Thinopyrum bessarabicum).
