Abstract

The base of research and programs breeding is the the Study of genetic diversity plant populations. One of the most important fruits in the world is Citrus sinensis that grows in tropical and semi-tropical climates. In Iran, some citrus varieties was traditionally cultivated, and recently their growing are increased, because of this Iran is the one of the 8 great country that produce citrus. Jiroft city has a tropical climate. It is third producer in Iran with 36000 hectare under cultivation citrus, and production 20-25 tons per hectare. Using molecular markers to identify genetic similarities and populations genetic studies is important and one of the most important molecular markers is Retrotransposons. The purpose of this study was to investigate genetic diversity of jiroft citrus sinensis cultivars using retrotransposon based molecular markers. DNA extraction was performed by the method of Doyle and Doyle. In this study 19 varieties of orange was consist of two domestic varieties and 17 imported varieties, that imported varieties are the most important commercial varieties in country, and Jiroft local varieties is one of the high yielding varieties suitable region. The cultivars were analyzed by 4 pair retrotransposon- based primers and PCR production were examined by agaros gel electrophoresis. Presence or absence of PCR production were scored and analyzed using multivariate methods. Mantel test indicated r=0.96 for the correlation between the similarity matrix and cophentic coefficient. Cluster analysis was performed by using UPGMA method and Dice similarity coefficient and cultivars were divided into 4 main groups.Overally, principal component analysis indicated that the first 8 components explained 88% of total variations. The first three components was justified 31%, 13% and 12% of the total variations respectively.
