Abstract

Wheat is a very strategic crop for many contries. Sunn pest is one of the most important pest of cereal particulary for wheat and barley. α-amylase enzyme catalyze the hydrolysis of the α-D- (1,4)- glucan linkage in starch components, glycogen and various other related carbohydrates. Plant α-amylase inhibitors show great potential as tools toengineer resistance of crop plants against pest. In the study the inhibitory effect of proteineous inhibitors isolated from three varieties of wheat (Triticum aestivum var. Alvand) and (Triticum turgidum lin. D6 , D18) was examined on gut α-amylase for sunn pest (Eurygaster integricepse). α-amylase inhibitors from wheat seeds was purified using Ion exchenge chromatography cloumm. Inhibition percentage and activity inhibition were recorded for the line D18 , D6 and Alvand variety (23.3%) 0.46 U/ml, (19.1%) 0.38 U/ml and (19.7%) 0.39 U/ml respectively. Enzyme electrophoresis bands were showed low enzyme activity in precence of inhibitor proteins in comparison with control (lack of inhibitor protein) in line of D18 than line D6 and Alvand variety and high inhibition activity was in line D18 to line D6 and Alvand variety
