Abstract

Hyssopus officinalis, is a plant of the minty family, and one of the valuable medicinal plants. This experiment was carried out in order to optimize the production of callus and cell suspension of Hyssop plant under glass cultivation conditions and the effect of the two chitosan and titanium oxide elicitor on the amount of phenol, Flavonoid and antioxidant compounds of Hyssop plant, was investigated. the experiments carried out in this study in two parts: consisted of the callus production of sample of leaf and stem and the second part of the elicitor usage on the cultivation of the resultant callus cell suspension in the form of factorial in a completely randomized design with three replication, was done. The callus regeneration experiment was carried out in the basic MS cultivation medium, plus various concentrations of the hormone 2,4-D (0, 0.5, 1, 1.5 mg/litre) using different samples (leaves and stem). The purpose of study in the first experiment was (induction of callus) contains of weight and diameter of callus and in second experiment was the amount of phenol, Flavonoid and antioxidant compounds. The results of the callus regeneration showed that the sample of the stem and hormone of 2,4-D in the concentration of 1.5 mg/litre for weight and callus diameter were the best treatment. The resultant of suspension cultivation showed that the highest wet weight and dry weight of foliaceous callus was related to chitosan 10 mg/litre, along with non-use of titanium nano oxide. In the leaf suspension, the maximum amount of phenol, Flavonoid and antioxidant compounds was observed in chitosan 20 with titanium nano dioxide 30, as well as chitosan 30 and titanium nano dioxide 90 mg. For the stem suspension the highest wet and dry weight callus, related to the witness treatment and , the maximum amount of phenol, Flavonoid and antioxidant compounds was related to the chitosan 30 mg/litre. Due to the results of the study, the chitosan usage of 30 mg/litre cause The increasing amound of phenol, Flavonoid and antioxidant compounds in the compounds in the cultivation conditions.
