Abstract
Colorectal cancer is one of the high prevalence disorders in the world which base of WHO has 3rd rank on deadliest diseases chart. On other hand, due to side effects of chemotherapy, surgery and radiotherapy, use of medication with plant base was on scientist’s spotlight in the field of oncology and molecular genetics. Secondary metabolite like Thymol, Carvacrol and Paracymene having antioxidant, anti-fungal and anti-microbial properties. The aim of this research is to evaluate the effect of extract of Oliveria decumbens Vent. on Bax and Bcl-2 genes expression on colorectal cancer. Primarily, extract of the plant obtained and by DPPH evaluated its antioxidant properties. MTT assay performed to find its cytotoxicity threshold on colorectal cancer. Results showing that 14.39 mg/ml of extract after 24 hrs can cause apoptosis on treated cancer cell line. Gene expression scrutiny done by real-time PCR and β-actin gene used for normalization. Results showing that Olivera decumbens Vent. extract upregulate the expression of Bax which is a pro-apoptosis gene and downregulate the expression of Bcl-2 gene which is an anti-apoptosis gene. At the end samples of aerial part of plant for De Novo transcriptome study has been send to Macrogen (South Korea) and the analysis of obtained data performed by bioinformatics softwares.
