Abstract

Lettuce is an annual leaf plant of Asteraceae family which is rich in minerals, vitamins and nutrients. The purpose of the present study was to evaluate the genetic diversity of 15 varieties and lettuce population using the ITS area. Sequences quality was evaluated using Chromas software and then clustered using ClustlW method by MegAlign software. phylogenic trees and matrix of differences and similarities’s sequences were compared. The results showed a low genetic diversity among cultivars and populations. According to the results of the ITS area about inseparability of lettuce populations based on geographical location and important traits, it can be concluded that the ITS is not very suitable for intrinsic genetic variation, but base on a comparison that accomplished between the lettuce used in this Research and rest of plants of the Asteraceae family (presented at the NCBI site), found out that ITS could be an appropriate tool for inter-species and inter-genus genetic assessments. The numerical value of the ratio (dN / dS) was 0.015, indicated a pure selection on the examined gene and not causing any main changes. On the other hand being under one number (dN \ dS) and number of few inserted and deleted places which (from 1551 positions, 376) indicate low variation between different lines, therefore, perhaps the inability of ITS to differentiate populations for this reason. Considering that the origin of the plants originates from the centers that are most diverse and because Varcanane varieties population is the most diverse, it is therefore necessary to collect the lettuce germplasm for breeding breeding in Varzaneh region Esfahan to pay more attention.
