Abstract

Cardiovascular disease is the most common cause of death in most countries of the world and is the most important factor in disability. Blood coagulation is also involved in the development of myocardial infarction and cerebral ischemia and embolism. Warfarin is a medicine that prevents blood coagulation. The proteins of CYP2C9, ORM1 and VKORC1 play a role in the effective dose of warfarin. In this study, the association of the genes encoding these proteins CYP2C9 *, CYP2C9 * 3, VKORC1 and ORM1 and the dose of warfarin was investigated. Genomic DNA was extracted from 50 blood samples. Genotyping was performed by CTPPPCR. The PT was measured by turbidometric method and the results were analyzed by SPSS software. PCR product from VKORC1 (rs9923231) containing 208bp and 384bp bands for normal GG homozygote polymorphism, 119 bp and 384bp bands for AA homogeneity polymorphism and 384bp, 208bp, and 119bp bands for heterozygote AG polymorphism Was obtained. For CYP2CP * 3 (rs1799853), bands of 387 bp and 125 bp express normal homozygote AA genotype, bands of 387 bp and 200 bp expressing homozygous mutant CC genotypes and bands of 387bp, 200bp and 125bp showed heterozygote. For CYP2C9 * 2 (rs1057910), bands of 406bp and 123 bp in the normal homozygote genotype CC and bands of 406bp, 221bp and 123bp indicated heterozygote CT. For ORM1 (rs17650), 404bp and 126bp bands express normal homozygote GG genotype and 404bp, 216bp and 123bp bands indicate heterozygote CT. VKORC1 (rs9923231) and CYP2C9 * 2 (rs1057910) with a significant amount of warfarin. Although the CYP2C9, VKORC1 and ORM1 genes may affect the dose required for warfarin, they are not the only factors that cause differentiation, but other factors are also involved in determining the dose required. These factors include genetic, nutritional or environmental factors that are specific to different ethnicities. By determining the dose of warfarin for each individual, based on environmental and genetic factors, the risk of bleeding and death at the onset of treatment decreases with warfarin.
