Abstract

Die-back caused by Paecilomyces variotii is one of destructive diseases of pistachio in kerman province. To determine disease agent, genetic variability assay and pathogenicity test of this species, sampling was randomly performed in five region of province. The isolated were cultured on PDA medium follow to purify by hyphal tip purification methods. Seventy isolated were purified and between these, twenty isolates were selected to next study. The isolated cultured on PDA medium showed yellow- brownish color with branching density phialids To assay pathogenicity test, Ohadi variety was used. Genetic variety was assayed using SSR marker by four pair specific primer of this genus. Symptoms of disease were note from 14, 21 and 28 days after inoculation. Data analyses by SPSS software showed that our isolated are related to five separate groups. The RE ،SW ٣ KS isolates showed high disease severity.SSR analysis and designing of hierarchical tree indicated that isolates with 41% coefficient similarity divided into 13 groups. Result of present study is illustrated by comparing the groupings progress related to the pathogenesis, areas sampled and genetic diversity, ther is not correlation between these parameter and so this fungal disease could not regarded as initial and principal agent of disease.
