Abstract

Diversity is the basis of all selections in plant breeding. This genetic selection requires diversity so that more genetic diversity will broaden its selection range. On the other hand, determining the features and classification of germplasm will enable breeders to avoid reworking while sampling of the population. Heterosis or hybrid superiority on parents’ mean depends on the genetic distance between them. Of course, to analyze the genetic distance between parents, it is necessary to classify the cultivars and the varieties. In this method, 15 Ajowan samples were prepared from the gene bank of the Bureau of Forests and Rangelands, Iran. It was conducted using RAPD short 10- nucleotide primers, which is one of PCR-based markers, in the Biotechnology and Genetic Engineering Lab in the Biotechnology Research Institute of Zabol University. Extraction of the plant samples was done based on Dellaporta method.To determine the quality and quantity of DNA, spectrophotometry and agarose gel were also used. The results showed that the total number of RAPD bands observed in 15 Ajowan samples was 50, where the Rafsanjan Ajowan samples (with 20 bands) and the Ardabil Ajowan samples (with 4 bands), respectively, had the highest and the lowest number. Moreover, cluster analysis based on Nei and Li and Jaccard similarity matrix and UPGMA method was performed. In both methods, the populations were divided into four sub- groups. The test showed a significantly high correlation between similarity matrices of Nei and Li and Jaccard methods (r= 0.99) indicating that both matrices provide the same information. Analysis into the main components of the rapid data showed that these special rates in the Nei and Li method for the first three components justify 62.3% of the total variance. On the other hand, loss of information into two or three main factors to explain the major features of the data was possible more easily, partly reflects the classification obtained from cluster analysis. It indicated that the majority of markers deliver independent information on the genetic structure of the Ajowan populations in this study. Given that only 8 Rapid primers could lead to this multiform in the Ajowan plant, this small amount of the primer provided a relatively acceptable solution, indicating that the RAPD markers, based on factors ranging from the conditions of experiments to the required testing time, costs, and material, can be useful tools to assess diversity, genetic similarity and to identify suitable areas for breeding.
